    The curvHDR Method for Gating Flow Cytometry Samples

    Motivation: High-throughput flow cytometry experiments produce hundreds of large multivariate samples of cellular characteristics. These samples require specialized processing to obtain clinically meaningful measurements. A major component of this processing is a form of cell subsetting known as gating. Manual gating is time-consuming and subjective. Good automatic and semi-automatic gating algorithms are very beneficial to high-throughput flow cytometry. Results: We develop a statistical procedure, named curvHDR, for automatic and semi-automatic gating. The method combines the notions of significant high negative curvature regions and highest density regions and has the ability to adapt well to human-perceived gates. The underlying principles apply to dimension of arbitrary size, although we focus on dimensions up to three. Accompanying software, compatible with contemporary flow cytometry informatics, is developed. Availability: Software for Bioconductor within R is available

    Reconstruction methods for acoustic particle detection in the deep sea using clusters of hydrophones

    This article focuses on techniques for acoustic noise reduction, signal filters and source reconstruction. For noise reduction, bandpass filters and cross correlations are found to be efficient and fast ways to improve the signal to noise ratio and identify a possible neutrino-induced acoustic signal. The reconstruction of the position of an acoustic point source in the sea is performed by using small-volume clusters of hydrophones (about 1 cubic meter) for direction reconstruction by a beamforming algorithm. The directional information from a number of such clusters allows for position reconstruction. The algorithms for data filtering, direction and position reconstruction are explained and demonstrated using simulated data.Comment: 7 pages, 13 figure

    Altered processing of food stimuli in adolescents with loss of control eating

    Loss of control eating (LOC) constitutes a common eating pathology in childhood and adolescence. Models developed for adult patients stress a biased processing of food-related stimuli as an important maintaining factor. To our knowledge, however, no EEG study to date investigated the processing of visual food stimuli in children or adolescents with LOC. Adolescents with at least one self-reported episode of LOC in the last four weeks and a matched control group completed a modified Go/NoGo task, with a numerical target or non-target stimulus being presented on one side of the screen and an irrelevant high-calorie food or neutral stimulus being presented on the opposite side. Mean P3 amplitudes were analyzed. In Go trials, the LOC group’s mean P3 amplitudes were comparable irrespective of distractor category, while for NoGo trials, mean P3 amplitudes were significantly higher when the distractor was a high-calorie food stimulus. This pattern was reversed in the control group. Results are interpreted in light of Gray’s reinforcement sensitivity theory. They might reflect altered processes of behavioral inhibition in adolescents with LOC upon confrontation with visual food stimuli

    Calibration sources are an indispensable tool for all detectors. In acoustic particle detection the goal of a calibration source is to mimic neutrino signatures as expected from hadronic cascades.     Progression of myopathology in Kearns-Sayre syndrome

    We report on the progression of myopathology by comparing two biopsies from a patient with a Kearns-Sayre-Syndrome. The first biopsy was taken in 1979 and showed 10% ragged-red fibers. Myopathic changes were slight including internal nuclei and fiber splitting in 10% of the fibers. Electron microscopy revealed typical mitochondrial abnormalities with regard to number and shape. In 1989 a second biopsy was performed for an extended analysis of mitochondrial DNA. This time less than 5% of all fibers were ragged-red. Severe myopathic changes could be detected which so far has rarely been reported in mitochondrial cytopathy

    Damage estimation of subterranean building constructions due to groundwater inundation – the GIS-based model approach GRUWAD

    The analysis and management of flood risk commonly focuses on surface water floods, because these types are often associated with high economic losses due to damage to buildings and settlements. The rising groundwater as a secondary effect of these floods induces additional damage, particularly in the basements of buildings. Mostly, these losses remain underestimated, because they are difficult to assess, especially for the entire building stock of flood-prone urban areas. For this purpose an appropriate methodology has been developed and lead to a groundwater damage simulation model named GRUWAD. The overall methodology combines various engineering and geoinformatic methods to calculate major damage processes by high groundwater levels. It considers a classification of buildings by building types, synthetic depth-damage functions for groundwater inundation as well as the results of a groundwater-flow model. The modular structure of this procedure can be adapted in the level of detail. Hence, the model allows damage calculations from the local to the regional scale. Among others it can be used to prepare risk maps, for ex-ante analysis of future risks, and to simulate the effects of mitigation measures. Therefore, the model is a multifarious tool for determining urban resilience with respect to high groundwater levels
